JAK2 Break Apart FISH Probe Kit
Oncology JAK2 Break Apart FISH Probe Kit Cat No CT-PAC174 Leukemia & Other Malignancies Diagnostic Kit The JAK2 Break Apart FISH Probe Kit is designed to detect rearrangements in the human JAK2 gene located on chromosome band 9p24.1. In addition to revealing breaks, which can lead to translocation of parts of the gene, inversion, or its fusion to other genes, the probe set can also be used to identify other JAK2 aberrations such as deletions or amplifications. Rearrangements and abnormal expression of the JAK2 gene – also known as THCYT3 or JTK10 – have been observed in acute myeloid and lymphoid leukemias and other malignancies. The probe is labeled with CytoOrange and CytoGreen. CytoOrange is a fluorophore with an excitation peak at 551nm and emission peak at 575nm, giving it an orange fluorescent signal with an appropriate filter; CytoGreen has an excitation peak at 495nm and emission peak at 518nm, giving it a green fluorescent signal with an appropriate filter.

The ZytoLight SPEC JAK2 Dual Color Break Apart Probe (PL248) is intended to be used for the qualitative detection of translocations involving the human JAK2 gene at 9p24.1 in cytologic specimens by fluorescence in situ hybridization (FISH). The probe is intended to be used in combination with the ZytoLight FISH-Cytology Implementation Kit (Prod. No. Z-2099-20). The product is intended for professional use only. All tests using the product should be performed in a certified, licensed anatomic pathology laboratory under the supervision of a pathologist/human geneticist by qualified personnel. The probe is intended to be used as an aid to the differential diagnosis of various cancers and therapeutic measures should not be initiated based on the test result alone.

Our JAK2 break apart probe is designed to detect JAK2 translocations. The probe comes labeled in orange and green, but can be customized to meet your needs.
Gene Background: JAK2 encodes a tyrosine kinase that directs progression of the JAK2/STAT signaling cascade, a pathway crucial to hematopoeisis and immune cell development.1 JAK2 translocations have been detected in a variety of myeloproliferative diseases and hematopoietic tumors.2 These rearrangements usually transfer JAK2’s 39 kinase domain to a fusion partner, resulting in constitutive activation of JAK2.3 These fusions are recurrent in myeloproliferative neoplasms (MPN), acute myeloid leukemia, acute lymphoblastic leukemia and lymphoma.2 Three fusion variants in MPN, namely PCM1-JAK2, BCR-JAK2 and ETV6-JAK2, are thought to be constitutively activated drivers of the disease process analogous to BCR-ABL1 in chronic myeloid leukemia.4
