How do I choose the right HPLC column?
So Step 1 – look in a column catalog. It’s gonna give you a guaranteed result.
OK let’s say that you don’t have a column catalog. Or let’s say that your chemical isn’t in one. Let’s say that you are sitting there saying, “Well Lee if I knew all of that I wouldn’t be asking you for advice.”
So that is a valid point. Let me take you to the next step.
In HPLC we have dozens of columns out there, but there is one column that dominates. If you look at all the column sales, about 85% of the column sales are reversed phase columns.
Why is that? To me that means that 85% of the separation problems must be solved by reversed phase. Not every one, 15% won’t be. But let’s play the odds.
What you want to do is to choose one really good reversed phase column. You want to buy the best base-deactivated C-18 column that you can get.
C-18 is going to be your most useful reversed phase column. There are dozens of different C-18 columns out there. What you want to find is the most modern C-18 column out there.
So here is a weird thing. When you look in the catalog they are going to list their oldest columns and their newest columns. And catalogs won’t really tell you that their old columns are lousy. That catalog – I hate to say it – is filled with lousy columns.
It doesn’t mean that Agilent makes lousy columns, it means that there are old-fashioned columns that they still make for old-fashioned applications.
If you are starting a new application you want to start with today’s best technology.
So my favorite column is eclipse plus made by Zorbax, by Agilent. There are have their SB-C18 that is also fantastic it is their #1 selling column. I personally like the Eclipse Plus.
But I won’t limit it to my Agilent friends. Waters make a couple of great columns. Waters SunFire, a beautiful column, a beautiful C18 base deactivated column. They also make one called XBridge. A different technology, very high-pH stable, an excellent column.
My friends at Phenomenex make a column called the Luna C18, the #1 selling silica in the world. There is a newer version called the Gemini C18 – I don’t like the cute names but I like the column. The Gemini C-18 is the latest and the greatest out of Phenomenex.
I feel bad for all the companies that I did not mention. There are other great ones out there.
But you start with a good base deactivated C18 column. My next piece of advice is you buy a column with the most efficiency you can get. We haven’t defined efficiency yet. Come back to one of my lectures and I will define efficiency for you. But it is a measure of the skinniness of the peak.
So when you are entering method development I have no idea how much efficiency that I need, so let’s start with the longest with the most efficiency. If that works and I have empty space I can go to a shorter column to save time. But if I start with a short column, it may not work and then I would have to go to a long column. So in other words, start with a really long, really high efficiency base-deactivated C18 column. In my world that would mean a 10 cm/1.8 micron, or a 15 cm/3.5 micron, or a 25 cm/5 micron – old fashioned!
Any of those are brute force columns.
So to summarize: good c18 base deactivated, high efficiency. Start with that. That one column will handle most of the applications out there in the reversed phase world.

Ultra strong basic compounds like amitriptyline can be eluted from the column in neutral pH values with excellent symmetrical peak shapes.

ProntoSIL C18 AQ C18 120/200 3/5/10 14 – 9 3 – 8 Used in aqueous mobile phases with an organic content below 10%.

ProntoSIL C18 AQ PLUS C18 120 3/5/10 17 3 – 8 ProntoSIL C18 AQ PLUS shows an enhanced stability at low pH’s down to pH 1. Also the packing shows excellent peak shapes in pure aqueous eluents but differs in shape selectivity compared to ProntoSIL C18 AQ. The application field for this support is mainly in combinatorial chemistry where the standard separation conditions are fast gradients from 0 – 100% organic and where the mobile phases include 0.1% TFA.

This packing shows an enhanced selectivity comparable to Lichrospher Select B from Merck.

Due to the bonding technology it shows an enhanced stability even at pH 1. The C4-packings show excellent properties for the separation of large biomolecules like proteins and peptides not only in the RP-mode but also in the HIC-mode.

ProntoSIL C1 C1 120 3/5 3 1 – 8 The C1 packing shows the lowest retention of the complete ProntoSIL line. The application area is mainly the separation of non-polar solutes. It can also be used for the separation of proteins in the HIC-mode. Due to the bonding technology the C1-bonding type is stable down to pH 1.

ProntoSIL C30 C30 120/200/300 3/5/10 25 – 13 1 – 8 ProntoSIL C30 is a stationary phase with a high carbon load. The high coverage of the support results in a very dense bonding and in an excellent shape selectivity and stability even at pH 1. Especially the wide pore supports are showing an enhanced shape selectivity. The application field of the C30 packings is the separation of isomers of carotenoids and other long alkyl chain solutes that can not be separated on classical C18 columns.

ProntoSIL Phenyl Phenyl 60/120 3/5 12 – 9.5 1 – 8 ProntoSIL Phenyl is a RP packing that offers different selectivities in comparison to brush type stationary phases like C18 or C8; it is fully endcapped. Due to the bonding technology it shows excellent stability even at pH 1.

ProntoSIL Amino NH2 120 3/5/10 4 3 – 8 This phase can be used in three modes: NP-mode, RP-mode and IC-mode. ProntoSIL Amino H NH2 120 5 4.5 3 – 8 This packing shows a different selectivity in Supercritical Fluid Chromatography (SFC). Due to a special bonding technology it shows a higher lifetime in carbohydrate analysis in the RP-mode.

ProntoSIL CN CN 120 3/5 6 – 5 3 – 8 In RP-mode, the application area of ProntoSIL CN is the separation of strong basic solutes. In NPmode, the ProntoSIL CN-support offers a complementary selectivity to the other NP-phases: Silica, Amino and DIOL. Due to the quick equilibration time of the CN-bonded phase it is the best choice for gradient elution in NPmode.

ProntoSIL OH DIOL 60/120 3/5/10 5.5 – 4 3 – 8 ProntoSIL OH is a DIOL bonded phase. The DIOL packing can be used in Normal Phase (NP) and Reversed Phase (RP) Chromatography. It is also suitable for Supercritical Fluid Chromatography (SFC). The silanophilic activity is lower than that of pure silica. This is particularly advantageous in Size Exclusion Chromatography (SEC). When the DIOL phase is employed in NP mode, the equilibration times are shorter than on a pure silica support, the selectivities, however, are similar.

The columns are pretested with a standard mixture described in EPA method 604/625.

ProntoSIL Enviro PTL proprietary 60 5 2.5 3 – 8 ProntoSIL Enviro PTL is a special stationary phase for the group determination of non-aromatic, monoaromatic, diaromatic and polyaromatic hydrocarbons in petroleum products. This stationary phase was designed to meet the requirements of European Norm prEN 12916 (June 1997). In this norm a resolution between Cyclohexane (non-aromatic) and o-Xylene (monoaromatic) of minimum 7 is required. Therefore the columns packed with this stationary phases are only offered in the length of 250 mm. The columns are pretested with a standard mixture described in prEN 12916.

This stationary phase was designed to meet the requirements of German industry norm DIN 38407 F12. In this procedure 17 pesticides have to be separated. ProntoSIL Enviro PES columns are able to separate this complex mixture in less than 45 minutes. The columns are shipped pretested with the pesticide standard mixture.

PolyEncap A C4 120/300 3/5/7/10 8 – 4 3 – 10 PolyEncap stationary phases are a family of bonded phases where the silica is polymeric encapsulated. The encapsulation is done by acrylic acid ester polymers which are additionally covalent bonded to the silica surface. This type of stationary phase offers different selectivities compared to classical bonded brush type stationary phases. PolyEncap A has a comparable selectivity to a brush type C8 packing.

Spheribond ODS 2 C18 80 3/5/10 12 3 – 8 ProntoSIL Spheribond ODS 2 is the recommended alternative for Waters Spherisorb ODS 2 . It is based on a type A silica and has the same physical chemical properties and bonding density like the original RP support. The selectivity of the two packings is the same. The typical application field of this classical packing with moderate silanophilic activity is pharmaceutical analytes. The Waters Spherisorb ODS 2 column is the most popular spherical silica support in pharmaceutical applications.
